Lyphilized ultra pure white powder needs to be reconstututed in PBS, Sodium Phosphate or RPMI and sterile Water supplemented with 0% glycerol or gelatin in as high as possible concentration aliquots to prevent post reconstitution degradation. Aliquoting with 25 mM HEPES-KOH, pH 7.2 and 1 mM DTT and 10% weight on volume glycerol is suitable for storage of the reconstituted powder. The solution can be stored for short term at +4 C 5 days and long term storage at - 85C. Freeze thaw cycles need to be omitted to maintain the protein activity. A typical loss of 15 % in each freeze thaw cycle can be expected.

Proteinase K s a broad-spectrum serine protease. The enzyme was discovered in 1974 in extracts of the fungus Engyodontium album (formerly Tritirachium album).Proteinase K is able to digest hair (keratin), hence, the name "Proteinase K". The predominant site of cleavage is the peptide bond adjacent to the carboxyl group of aliphatic and aromatic amino acids with blocked alpha amino groups. It is commonly used for its broad specificity. This enzyme belongs to Peptidase family S8. The molecular weight of Proteinase K is 28,900 Daltons (28.9 kDa).
