BackgroundNCK1 (NCK alpha) is a cytoplasmic adaptor protein that plays a universal role in coordinating the signaling networks critical for organizing the actin cytoskeleton, cell movement, or axon guidance, connecting transmembrane receptors to multiple intracellular signaling pathways. It contains one SH2 domain, through which NCK1 binds to phosphorylated domains of transmembrane signaling moleculs or certain adaptor proteins, and three SH3 domains for binding proline-rich sequences of other molecules involved in the process of nucleation and polymerization of the actin cytoskeleton.
