ELISA kit description
GENTAUR's human ADAMTS4 ELISA test is a ready to use ELISA kit. We use a monoclonal from mouse specific for ADAMTS4 was used as capure antibodies on the ELISA test plate. The internal standards (CHO, F213-C685) and your samples are measured with a biotin conjugated polyclonal from goat specific for ADAMTS4 will than be washed with PBS. The colour reaction with TMB on HRP will colour the well from blue to yellow with the stop solution. The more human ADAMTS4 in your sample the higher the ELISA signal will be compared to this elisa's standard curve.

How the elisa works
Sample the amount of 100ul/well of the standard solution at 40000pg/ml, 20000pg/ml, 10000pg/ml, 5000pg/ml, 2500pg/ml, 1250pg/ml, 625pg/ml human ADAMTS4 standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffers into the control well (Zero well). Add 0.1ml of each properly diluted sample of human cell culture supernatant, serum or plasma(heparin, EDTA) to the ELISA plate. Keep in mind that each human ADAMTS4 standard solution and each sample be measured in duplicate.

ADAMTS4, A disintegrin and metalloproteinase with thrombospondin motifs 4 is an enzyme that in humans is encoded by the ADAMTS4 gene. ADAMTS4 is a member of the large ADAMTS family of zinc-dependent proteases. The human ADAMTS4 gene is mapped to chromosome 1 by somatic cell hybrid analysis. The enzyme encoded by this gene lacks a C-terminal TS motif. It is responsible for the degradation of aggrecan, a major proteoglycan of cartilage, and brevican, a brain-specific extracellular matrix protein. The cleavage of aggrecan and brevican suggests key roles of this enzyme in arthritic disease and in the central nervous system, potentially, in the progression of glioma.
