    Tumor Necrosis Factor alpha is a cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia. Under certain conditions it can stimulate cell proliferation and induce cell differentiation._x000B_Genetic variations in TNF both influence susceptibility to hepatitis B virus (HBV) infection and are involved in susceptibility to malaria. Additionally, genetic variations in TNF are a cause of susceptibility psoriatic arthritis (PSORAS). PSORAS is an inflammatory, seronegative arthritis associated with psoriasis. It is a heterogeneous disorder ranging from a mild, non-destructive disease to a severe, progressive, erosive arthropathy. Five types of psoriatic arthritis have been defined: asymmetrical oligoarthritis characterized by primary involvement of the small joints of the fingers or toes; asymmetrical arthritis which involves the joints of the extremities; symmetrical polyarthritis characterized by a rheumatoidlike pattern that can involve hands, wrists, ankles, and feet; arthritis mutilans, which is a rare but deforming and destructive condition; and arthritis of the sacroiliac joints and spine (psoriatic spondylitis).

    ELISA: This antibody has been used as a coating antibody at 5µg/ml in a two site ELISA with biotin conjugated TNF antibody (Cat. No. L190M). Typically the ELISA has a working range of 0-10000 pg/ml and a sensitivity of 5 pg/ml with the appropriate standard. Recommended sample volumes are 25-500 µl. Recovery of hTNF in human serum is typically >85% in the ELISA. IC: This antibody has been used to immunoprecipitate > 90% TNF in solution using a double-antibody method (Midgley and Hepburn).

    The Mouse anti Tumor Necrosis Factor alpha (TNF )* is a α- or alpha protein sometimes glycoprotein present in blood. This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided. Aplha, transcription related growth factors and stimulating factors or repressing nuclear factors are complex subunits of proteins involved in cell differentiation. Complex subunit associated factors are involved in hybridoma growth, Eosinohils, eritroid proliferation and derived from promotor binding stimulating subunits on the DNA binding complex. NFKB 105 subunit for example is a polypetide gene enhancer of genes in B cells.
