Long description
The CD3 epitope is expressed on the epsilon chain of the CD3/T cell antigen receptor (TcR) complex. CD3 is present on 65-85% of thymocytes and has a mitogenic effect on peripheral blood T cells. CD3 indentifies human T cells expressing the 22-28,000 M.W. surface antigen. CD8 identifies cytotoxic/suppressor human T cells expressing the 32 and 43,000 M.W. surface antigen. CD45 identifies all human leukocytes, lymphocytes, monocytes, polymorphonuclear cells, eosinphils and basophils in peripheral blood expressing the 180-220K M.W. T200 family surface antigen. CD45 is also present on thymus,spleen, tonsil and progenitor cells in bone marrow.

Antibody s suited for
PBMC: Add 10 µl of 3-ColorTM antibody reagent/10^6 PBMC in 100 µl PBS. Mix gently and incubate for 15 minutes at 2 ¡ to 8¡ C. Wash twice with PBS. Wash twice with PBS or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. WHOLE BLOOD: Add 10 µl of Exalpha Biological's 3-ColorTM antibody reagent MAB/100 µl of whole blood. Mix gently and incubate for 15 minutes at room temperature 20 ¡ C. Lyse the whole blood. Wash once with PBS and analyze or fix with 0.5% v/v of paraformaldehyde in PBS and analyze. See instrument manufacturerÕs instructions for Lysed Whole Blood and Immunofluorescence analysis with a flow cytometer or microscope.

Description
This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided. Cytotoxic T cells cluster differentiators for flow cytometry. This transmembrane glycoprotein is detected by monoclonals and has a lot of clones available with different affinity.

Properties
If you buy Antibodies supplied by nordc they should be stored frozen at - 24°C for long term storage and for short term at + 5°C. Cy5 antibodies are excited by the 650-nanometer wave of an argon laser and detected at a 670-nanometer captor. This nordc Fluorescein isothiocyanate (FITC) antibody is currently after some BD antibodies the most commonly used fluorescent dye for FACS. When excited at 488 nanometers, FITC has a green emission that's usually collected at 530 nanometers, the FL1 detector of a FACSCalibur or FACScan. FITC has a high quantum yield (efficiency of energy transfer from absorption to emission fluorescence) and approximately half of the absorbed photons are emitted as fluorescent light. For fluorescent microscopy applications, the 1 FITC is seldom used as it photo bleaches rather quickly though in flow cytometry applications, its photo bleaching effects are not observed due to a very brief interaction at the laser intercept. nordc FITC is highly sensitive to pH extremes.
