  • Gene info
There are four distinct ligase proteins found in mammalian cells. The activity of these DNA ligases is required for the joining of DNA strand breaks that occur during DNA replication, repair and recombination. DNA Ligase III gene product migrates as a 100 kDa (a-form) protein in somatic tissues, is ubiquitously expressed and complexes with XRCC-1 and a 97 kDa (-form) protein that is found in testis. DNA Ligase III is involved in rejoining DNA single-strand breaks and in the base excision repair pathway.

  • Concept
Scope note: A DNA amplification technique based upon the ligation of OLIGONUCLEOTIDE PROBES. The probes are designed to exactly match two adjacent sequences of a specific target DNA. The chain reaction is repeated in three steps in the presence of excess probe: (1) heat denaturation of double-stranded DNA, (2) annealing of probes to target DNA, and (3) joining of the probes by thermostable DNA ligase. After the reaction is repeated for 20-30 cycles the production of ligated probe is measured.
