GENTAUR's human Adiponectin ELISA test is a ready to use ELISA kit. We use a monoclonal from mouse specific for Adiponectin was used as capure antibodies on the ELISA test plate. The internal standards (NSO, E19-N244) and your samples are measured with a biotin conjugated polyclonal from goat specific for Adiponectin will than be washed with PBS. The colour reaction with TMB on HRP will colour the well from blue to yellow with the stop solution. The more human Adiponectin in your sample the higher the ELISA signal will be compared to this elisa's standard curve.

How the elisa works
Sample the amount of 100ul/well of the standard solution at 100ng/ml, 50ng/ml, 25ng/ml, 12.5ng/ml, 6.25ng/ml, 3.12ng/ml, 1.56ng/ml human Adiponectin standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffers into the control well (Zero well). Add 0.1ml of each properly diluted sample of human cell culture supernatant, serum, plasma(heparin, EDTA), tissue homogenates or urine to the ELISA plate. Keep in mind that each human Adiponectin standard solution and each sample be measured in duplicate.

Adiponectin(ADPN) is a hormone secreted by adipocytes that regulates energy homeostasis and glucose and lipid metabolism. Adiponectin is a new member of the family of soluble defense collagens, in hematopoiesis and immune responses. It is an important negative regulator in hematopoiesis and immune systems and raise the possibility that it may be involved in ending inflammatory responses through its inhibitory functions. Adiponectin is mapped to 3q27 and can protect the organism from systemic inflammation by promoting the clearance of early apoptotic cells by macrophages through a receptor-dependent pathway involving calreticulin. The standard product used in this kit is the product of gene recombination, consisting of 226(19-244) amino acids with the molecular mass of 36KDa after glycosylation.
