Long description
Mucins are the major glycoprotein component of mucous and their function is to protect epithelia from adverse conditions such as dehydration, infection, physical and chemical injury. They are large glycoproteins which vary in size and exist as either membrane bound or secreted molecules. The secreted molecules are subdivided into gel-forming and non gel-forming mucins. Mucin 5AC (MUC5AC) is a high molecular weight gel-forming glycoprotein, which is primarily expressed in the goblet cells of the respiratory epithelium, gastric epithelium and reproductive mucosa. The aberrant expression of MUC5AC has been reported in some cancers, including gastric, colon, endometrial adenocarcinomas and pancreatic cancer, suggesting a potential role for MUC5AC as a marker of these neoplasia. Increased levels of MUC5AC have also been reported in some pathological conditions such as asthma and cystic fibrosis.

45M1 is a mouse monoclonal IgG1 antibody derived by fusion of mouse myeloma cells with spleen cells from a mouse immunized with M1 mucin purified from fluid taken from an ovarian mucinous cyst.

Each vial contains 500 µl 0.2 mg/ml purified monoclonal antibody in phosphated buffered saline (PBS) containing 50% glycerol + 0.2% bovine serum albumin (BSA) + 0.02% sodium azide.

Antibody s specificity
The 45M1 antibody is directed against human mucin 5AC and has been reported to cross-react with avian, mouse, rat, hamster, guinea-pig , porcine, and canine mucin 5AC. The 45M1 antibody is reported to react specifically with an epitope located in the N-terminal region of the C-terminal cysteine-rich part of MUC5AC.

Antibody s suited for
The 45M1 antibody is suitable for the detection of mucin 5AC by ELISA, flow cytometry, western blotting and immunoprecipitation. The antibody is also suitable for the detection of MUC5AC by immunocytochemistry and immunohistochemistry on frozen tissues. Staining of paraffin embedded tissues with 45M1 is possible following heat mediated antigen retrieval of paraffin sections using citrate buffer. Optimal antibody dilutions for the different applications should be determined by titration.

Description
This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided.

Test
Mouse or mice from the Mus musculus species are used for production of mouse monoclonal antibodies or mabs and as research model for humans in your lab. Mouse are mature after 40 days for females and 55 days for males. The female mice are pregnant only 20 days and can give birth to 10 litters of 6-8 mice a year. Transgenic, knock-out, congenic and inbread strains are known for C57BL/6, A/J, BALB/c, SCID while the CD-1 is outbred as strain.
