    This standard is prepared by pooling serum obtained from a large number of qualified (healthy, nonmedicated) adult blood donors. IgG1-- 5.57 mg/ml. IgG2--3.08 mg/ml. IgG3--0.39 mg/ml. IgG4-- 0.55 mg/ml. Total IgG--9.55 mg/ml. The determination of the concentration of each of the four subclasses was carried out by single radial immunodiffusion (RID, Mancini) in agar gel Nordic Nr. 2 using subclass-specific polyclonal antisera and the WHO reference serum 67/97 as the primary standard. The Who primary standard and the NOR-01 secondary standard were compared using them as calibrators for the investigations of the IgG subclasses content in a set of sera comprising he primary standard, the secondary standard and the sera of patients covering a large range of concentrations for each of the subclasses. The assay techniques used were ELISA and the dot-immunobinding assay (DIBA) with Nordimmune purified monoclonal antibodies to each of the four subclasses, and RID with polyclonal antisera. Monoclonal antibodies of the following murine hybridoma clones were used: Anti IgG1--NI132--(HP 6186). Anti IgG2-- NI 6014--(HP 6014). Anti IgG3--NI 6050--(HP 6050). Anti IgG3m(u)--NI 86--(HP 6080). Anti IgG4--NI 315-- (HP 6206). The subclass specificity of theses MAbs had been tested extensively in various assay techniques and was confirmed by ELISA and a Western blotting technique. Egression analysis was performed for data obtained by ELISA versus DIBA, and RID versus DIBA. The results showed a good correlation between DIBA an ELISA irrespective of the IgG subclass. When DIBA and RID were compared, there was also a significant correlation between the measurement of the IgG subclasses in these two ways. To estimate the inter-assay variation, the secondary standard NOR-01, the WHO primary standard and an additional pool of normal human sera were screened several times on various days using DIBA as the most sensitive of the methods used in the characterization of the secondary standard NOR-01. A significant correlation was obtained between the subclass values ascribed to the standard serum NOR-01 and those stated for the WHO reference serum. Irrespective of the IgG subclass measured, a coefficient of variation of 10% or less was obtained. The use of two different monoclonal antibodies for the measurement of IGG3 (one isotype and one allotype-specific), produced almost identical results. The apparent detection limit in the DIBA method was established to be in the range of 1-10 ng, also independent of the subclass measured. Human standard serum NOR-01 has been tested and found negative for HbsAg and antibodies to HIV (HTLVIII). Human standard serum NOR-01 is intended for use as a calibrator to prepare standard curves in quantitative determinations of human IgG subclasses.

    The product is shipped at ambient temperature. After arrival store, divided into small aliquots, frozen at or below -20°C. Working dilutions may be prepared by adding the required amount of phosphate buffered saline (PBS) containing 0.05% Tween and 1% (w/v) bovine serum albumin, should not be refrozen and preferably used within 24 hours. Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation and will not affect the performance of the product.
