  • Long description
    The c-kit proto-oncogene encodes a transmembrane receptor with tyrosine kinase activity, c-kit (CD117), also known as stem cell factor receptor (SCFR), which is closely related to the platelet-derived growth factor receptor family. The c-kit oncoprotein plays a role during normal hematopoiesis, gametogenesis and melanogenesis. It is expressed in among others normal breast epithelium, melanocytes, mast cells and glial cells. Defects in c-kit are a cause of several diseases, such as gastrointestinal stromal tumor (GIST), and testicular germ cell tumor. Also acute myelogenous leukemia (AML), a malignant disease in which hematopoietic precursors are arrested in an early stage of development, is related to mutation in c-kit. As a result, the antibody to CD117 is suitable the characterization of certain leukemias.

    T595 is a mouse monoclonal IgG1 antibody derived by fusion of p3-NS1-Ag4-1 mouse myeloma cells with spleen cells from a BALB/c mouse immunized with prokaryotic recombinant protein corresponding to the three N-terminal C2-like extracellular domains of the human c-kit oncoprotein.

  • Antibody s suited for
    The T595 antibody is suitable for the detection of the c-kit protein by immunocytochemistry on isolated cells, and immunohistochemistry on frozen and formalin fixed paraffin embedded tissues. For the latter application the high temperature antigen unmasking technique with 1 mM EDTA (pH 8.0) should be used (two cycles of 10 minutes in a microwave oven at 500 W ; see reference 5), and incubation with the primary antibody for 60 min. at 25 °C. Optimal antibody dilutions for the different applications should be determined by titration; recommended dilution 1:20 -1:40.

    Store undiluted antibody at 2-8ºC (until expiry date). For prolonged storage: store aliquots at -20ºC.

    This antibody needs to be stored at + 4°C in a fridge short term in a concentrated dilution. Freeze thaw will destroy a percentage in every cycle and should be avoided. Antibody against the Hu protein or peptide or recombinant supplied in 1 volumes. Ask for quote if you need bulk.
