BackgroundGrowth Differentiation Factor 15 (GDF-15), also known as Macrophage Inhibitory Cytokine-1 (MIC-1), belongs to the superfamily of transforming growth factor-beta (TGF-β). During normal conditions, GDF-15 is expressed at low levels in organs such as kidney, stomach, bladder, and placenta. Under stress conditions, the expression is upregulated by various stress-related proteins such as IL-1β, TNF-α, IL-2 in a tissue-specific manner. Elevated expressions are observed in diseased states such as cancer, inflammation, cardiovascular disease, and kidney disease. Hence, GDF-15 serves as an efficient biomarker for cancer and cardiovascular disease. BioVision’s GDF-15 (Human) ELISA kit is based on the Sandwich ELISA principle to quantitatively measure the amount of GDF-15 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and subsequently washed with Wash Buffer. The HRP-Streptavidin is added, and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is proportional to the amount of GDF-15 in the sample or standard.

ConceptScope note: An immunoassay utilizing an antibody labeled with an enzyme marker such as horseradish peroxidase. While either the enzyme or the antibody is bound to an immunosorbent substrate, they both retain their biologic activity; the change in enzyme activity as a result of the enzyme-antibody-antigen reaction is proportional to the concentration of the antigen and can be measured spectrophotometrically or with the naked eye.
